    Modeling columnar thin films as platforms for surface-plasmonic-polaritonic optical sensing

    Via exploitation of surface plasmon polaritons (SPPs), columnar thin films (CTFs) are attractive potential platforms for optical sensing as their relative permittivity dyadic and porosity can be tailored to order. Nanoscale model parameters of a CTF were determined from its measured relative permittivity dyadic, after inverting the Bruggeman homogenization formalism. These model parameters were then used to determine the relative permittivity dyadic of a fluid-infiltrated CTF. Two boundary-value problems were next solved: the first relating to SPP-wave propagation guided by the planar interface of a semi-infinitely thick metal and a semi-infinitely thick CTF, and the second to the plane-wave response of the planar interface of a finitely thick metallic layer and a CTF in a modified Kretschmann configuration. Numerical studies revealed that SPP waves propagate at a lower phase speed and with a shorter propagation length, if the fluid has a larger refractive index. Furthermore, the angle of incidence required to excite an SPP wave in a modified Kretschmann configuration increases as the refractive index of the fluid increases

    On chemiluminescent emission from an infiltrated chiral sculptured thin film

    The theory describing the far-field emission from a dipole source embedded inside a chiral sculptured thin film (CSTF), based on a spectral Green function formalism, was further developed to allow for infiltration of the void regions of the CSTF by a fluid. In doing so, the extended Bruggeman homogenization formalism--which accommodates constituent particles that are small compared to wavelength but not vanishingly small--was used to estimate the relative permittivity parameters of the infiltrated CSTF. For a numerical example, we found that left circularly polarized (LCP) light was preferentially emitted through one face of the CSTF while right circularly polarized (RCP) light was preferentially emitted through the opposite face, at wavelengths within the Bragg regime. The centre wavelength for the preferential emission of LCP/RCP light was red shifted as the refractive index of the infiltrating fluid increased from unity, and this red shift was accentuated when the size of the constituent particles in our homogenization model was increased. Also, the bandwidth of the preferential LCP/RCP emission regime decreased as the refractive index of the infiltrating fluid increased from unity

    Hybrid Core-Shell Scaffolds for Bone Tissue Engineering

    The tissue engineering applications of coaxial electrospinning are growing due to the potential increased functionality of the fibres compared to basic electrospinning. Previous studies of core and shell scaffolds have placed the active elements in the core, however, the surface response to a biomaterial affects the subsequent behaviour, thus here hydroxyapatite (HA) was added to the shell. Coaxial electrospun polycaprolactone (PCL)-polylactic acid (PLA)/HA (core-shell) scaffolds were produced in 2D sheets using a plate collector, or 3D tubes for bone tissue engineering using a rotating needle collector. The scaffolds include high hydroxyapatite content while retaining their structural and mechanical integrity. The effect of the collector type on fibre diameter, fibre alignment and mechanical properties have been evaluated, and the impact of HA incorporation on bioactivity, BMP-2 release, cell behaviour and mechanical properties for up to 12 weeks degradation were assessed. Fibre uniformity in coaxial electrospinning depends on the relative flow rate of the core and shell solutions. Using a rotating needle collector increased fibre alignment compared to a stationary collector, without affecting fibre diameter significantly, while HA content increased fibre non-uniformity. Coaxial PCL-PLA/HA fibres exhibited significantly higher bioactivity compared to PCL-PLA scaffolds due to the surface exposure of the HA particles. Apatite formation increased with increasing SBF immersion time. Coaxial tubular scaffolds with and without HA incorporation showed gradual reductions in their mechanical properties over 12 weeks in PBS or SBF but still retained their structural integrity. Coaxial scaffolds with and without HA exhibited gradual and sustained BMP-2 release and supported MSCs proliferation and differentiation with no significant difference between the two scaffolds types. These materials therefore show potential applications as bone tissue engineering scaffolds

    Using environmental engineering to increase hand hygiene compliance: a cross-over study protocol

    Introduction Compliance with hand hygiene recommendations in hospital is typically less than 50%. Such low compliance inevitably contributes to hospitalacquired infections that negatively affect patients’ wellbeing and hospitals’ finances. The design of the present study is predicated on the assumption that most people who fail to clean their hands are not doing so intentionally, they just forget. The present study will test whether psychological priming can be used to increase the number of people who clean their hands on entering a ward. Here, we present the protocol for this study. Methods and analysis The study will use a randomised cross-over design. During the study, each of four wards will be observed during four conditions: olfactory prime, visual prime, both primes and neither prime. Each condition will be experienced for 42 days followed by a 7-day washout period (total duration of trial=189 days). We will record the number of people who enter each ward and whether they clean their hands during observation sessions, the amount of cleaning material used from the dispensers each week and the number of hospital-acquired infections that occur in each period. The outcomes will be compared using a regression analysis. Following the initial trail, the most effective priming condition will be rolled out for 3months in all the wards.     On columnar thin films as platforms for surface-plasmonic-polaritonic optical sensing: higher-order considerations

    The ability to tailor the porosity and optical properties of columnar thin films (CTFs) renders them promising platforms for optical sensing. In particular, surface-plasmon-polariton (SPP) waves, guided by the planar interface of an infiltrated CTF and a thin layer of metal, may be harnessed to detect substances that penetrate the void regions in between the columns of a CTF. This scenario was investigated theoretically using a higher-order homogenization technique, based on an extended version of the second-order strong-permittivity-fluctuation theory, which takes into account the size of the component particles which make up the infiltrated CTF and the statistical distribution of these particles. Our numerical studies revealed that as the size of the component particles increases and as the correlation length that characterizes their distribution increases: (i) the phase speed of the SPP wave decreases and the SPP wave's attenuation increases; (ii) the SPP wave's penetration into the CTF decreases; (iii) the angle of incidence required to excite the SPP wave in a modified Kretschmann configuration increases; (iv) the sharpness of the SPP trough in the graph of reflectance versus angle of incidence increases; and (v) the sensitivity to changes in refractive index of the infiltrating fluid decreases

    The use of nanovibration to discover specific and potent bioactive metabolites that stimulate osteogenic differentiation in mesenchymal stem cells

    Bioactive metabolites have wide-ranging biological activities and are a potential source of future research and therapeutic tools. Here, we use nanovibrational stimulation to induce osteogenic differentiation of mesenchymal stem cells, in the absence of off-target, nonosteogenic differentiation. We show that this differentiation method, which does not rely on the addition of exogenous growth factors to culture media, provides an artifact-free approach to identifying bioactive metabolites that specifically and potently induce osteogenesis. We first identify a highly specific metabolite, cholesterol sulfate, an endogenous steroid. Next, a screen of other small molecules with a similar steroid scaffold identified fludrocortisone acetate with both specific and highly potent osteogenic-inducing activity. Further, we implicate cytoskeletal contractility as a measure of osteogenic potency and cell stiffness as a measure of specificity. These findings demonstrate that physical principles can be used to identify bioactive metabolites and then enable optimization of metabolite potency can be optimized by examining structure-function relationships

    Short-term prediction of threatening and violent behaviour in an Acute Psychiatric Intensive Care Unit based on patient and environment characteristics

    <p>Abstract</p> <p>Background</p> <p>The aims of the present study were to investigate clinically relevant patient and environment-related predictive factors for threats and violent incidents the first three days in a PICU population based on evaluations done at admittance.</p> <p>Methods</p> <p>In 2000 and 2001 all 118 consecutive patients were assessed at admittance to a Psychiatric Intensive Care Unit (PICU). Patient-related conditions as actuarial data from present admission, global clinical evaluations by physician at admittance and clinical nurses first day, a single rating with an observer rated scale scoring behaviours that predict short-term violence in psychiatric inpatients (The Brøset Violence Checklist (BVC)) at admittance, and environment-related conditions as use of segregation or not were related to the outcome measure Staff Observation Aggression Scale-Revised (SOAS-R). A multiple logistic regression analysis with SOAS-R as outcome variable was performed.</p> <p>Results</p> <p>The global clinical evaluations and the BVC were effective and more suitable than actuarial data in predicting short-term aggression. The use of segregation reduced the number of SOAS-R incidents.</p> <p>Conclusions</p> <p>In a naturalistic group of patients in a PICU segregation of patients lowers the number of aggressive and threatening incidents.     Search for dark matter produced in association with bottom or top quarks in √s = 13 TeV pp collisions with the ATLAS detector

    A search for weakly interacting massive particle dark matter produced in association with bottom or top quarks is presented. Final states containing third-generation quarks and miss- ing transverse momentum are considered. The analysis uses 36.1 fb−1 of proton–proton collision data recorded by the ATLAS experiment at √s = 13 TeV in 2015 and 2016. No significant excess of events above the estimated backgrounds is observed. The results are in- terpreted in the framework of simplified models of spin-0 dark-matter mediators. For colour- neutral spin-0 mediators produced in association with top quarks and decaying into a pair of dark-matter particles, mediator masses below 50 GeV are excluded assuming a dark-matter candidate mass of 1 GeV and unitary couplings. For scalar and pseudoscalar mediators produced in association with bottom quarks, the search sets limits on the production cross- section of 300 times the predicted rate for mediators with masses between 10 and 50 GeV and assuming a dark-matter mass of 1 GeV and unitary coupling. Constraints on colour- charged scalar simplified models are also presented. Assuming a dark-matter particle mass of 35 GeV, mediator particles with mass below 1.1 TeV are excluded for couplings yielding a dark-matter relic density consistent with measurements

    The evolution of lung cancer and impact of subclonal selection in TRACERx

    Lung cancer is the leading cause of cancer-associated mortality worldwide. Here we analysed 1,644 tumour regions sampled at surgery or during follow-up from the first 421 patients with non-small cell lung cancer prospectively enrolled into the TRACERx study. This project aims to decipher lung cancer evolution and address the primary study endpoint: determining the relationship between intratumour heterogeneity and clinical outcome. In lung adenocarcinoma, mutations in 22 out of 40 common cancer genes were under significant subclonal selection, including classical tumour initiators such as TP53 and KRAS. We defined evolutionary dependencies between drivers, mutational processes and whole genome doubling (WGD) events. Despite patients having a history of smoking, 8% of lung adenocarcinomas lacked evidence of tobacco-induced mutagenesis. These tumours also had similar detection rates for EGFR mutations and for RET, ROS1, ALK and MET oncogenic isoforms compared with tumours in never-smokers, which suggests that they have a similar aetiology and pathogenesis. Large subclonal expansions were associated with positive subclonal selection. Patients with tumours harbouring recent subclonal expansions, on the terminus of a phylogenetic branch, had significantly shorter disease-free survival. Subclonal WGD was detected in 19% of tumours, and 10% of tumours harboured multiple subclonal WGDs in parallel. Subclonal, but not truncal, WGD was associated with shorter disease-free survival. Copy number heterogeneity was associated with extrathoracic relapse within 1 year after surgery. These data demonstrate the importance of clonal expansion, WGD and copy number instability in determining the timing and patterns of relapse in non-small cell lung cancer and provide a comprehensive clinical cancer evolutionary data resource
